The MWC model, proposed more than 50 years ago, is elegantly simple yet powerful in predicting the behavior of allosteric proteins like hemoglobin. Its prediction power for ion channel gating has been beautifully demonstrated in the studies of BK channels. Our present work aims to determine how accurate the MWC model predicts TRPV1 activation induced by vanilloids. Our findings support the notion that the evolutionary drive upon allosteric proteins applies generally to multi-subunit proteins.
