Herein, we designed and developed a single two-photon ratiometric fluorescence probe (TMF2P) for selective and accurate determination of mitochondrial MAO-A in live neurons. It was discovered that the increases in MAO-A levels under oxidative stress resulted in an elevated influx of Ca 2+ flow into mitochondria through the transient receptor potential melastatin 2 (TRPM2) channels.
