Using whole genome sequencing and conventional cytogenetics, we discovered that CRISPR-Cas9 cuts led to genomic instability, including chromosomal rearrangements and polyploidization, and ultimately to delayed cell death, 2-3 weeks after the induction of CRISPR-Cas9 DSBs.
